چکیده
ABSTRACT Acinetobacter baumannii possesses a single divergent luxR / luxRI -type quorum-sensing (QS) locus named abaR abaI . This also contains third gene located between and , which we term abaM that codes for an uncharacterized member of the RsaM protein family known to regulate N -acylhomoserine lactone (AHL)-dependent QS in other beta- gammaproteobacteria. Here, show disruption via T26 insertion A. strain AB5075 resulted increased production -(3-hydroxydodecanoyl)- l -homoserine enhanced surface motility biofilm formation. In contrast wild type ::T26 mutant, virulence mutant was completely attenuated Galleria mellonella infection model. Transcriptomic analysis revealed AbaM differentially regulates at least 76 genes, including csu pilus operon acinetin 505 lipopeptide biosynthetic operon, are involved adherence, formation virulence. A comparison type, transcriptomes, indicates ?21% regulon operon. Moreover, genes ( ) were among most upregulated mutant. lux -based reporter fusions expression is positively regulated by but negatively autoregulated. Overall, data presented this work demonstrates plays central role regulating QS, virulence, motility, IMPORTANCE multiantibiotic-resistant pathogen global health care importance. Understanding regulation could aid development novel anti-infective strategies. code receptor synthase components (AHL) lactone-dependent quorum sensing system separated although mutation AHL production, development, it attenuation found control both QS-dependent QS-independent genes. The significance lies identification AbaM, ortholog plant pathogens, as modulator human pathogen.

Quorum Sensing in Microbial Virulence
Cell-to cell communication occurs via a signaling pathway referred to as quorum sensing. There are four main types of these systems according to the chemical nature of signal molecules used by microorganisms to elicit expression of target genes in response to environmental stimuli or need of microbial communities. Type I system acts by using acyl homoserine lactones as signals to trigger the ex...
متن کاملBacteria that inhibit quorum sensing decrease biofilm formation and virulence in Pseudomonas aeruginosa PAO1.
In this study, we investigated the biotherapeutic potential of previously isolated quorum quenching (QQ) bacteria. Some of them produce and secrete small compounds that inhibit quorum sensing (QS), others quench QS by enzymatic degradation of N-acylhomoserine lactones (AHLs). The supernatant of cultures of these isolates was tested for inhibitory properties against P. aeruginosa PAO1 biofilms. ...
متن کاملRole of the luxS quorum-sensing system in biofilm formation and virulence of Staphylococcus epidermidis.
Nosocomial infections caused by Staphylococcus epidermidis are characterized by biofilm formation on implanted medical devices. Quorum-sensing regulation plays a major role in the biofilm development of many bacterial pathogens. Here, we describe luxS, a quorum-sensing system in staphylococci that has a significant impact on biofilm development and virulence. We constructed an isogenic Deltalux...
متن کاملTwo quorum sensing systems control biofilm formation and virulence in members of the Burkholderia cepacia complex
The Burkholderia cepacia complex (Bcc) consists of 17 closely related species that are problematic opportunistic bacterial pathogens for cystic fibrosis patients and immunocompromised individuals. These bacteria are capable of utilizing two different chemical languages: N-acyl homoserine lactones (AHLs) and cis-2-unsaturated fatty acids. Here we summarize the current knowledge of the underlying...
متن کاملEffect of Cinnamon Oil on Quorum Sensing-Controlled Virulence Factors and Biofilm Formation in Pseudomonas aeruginosa
Quorum sensing (QS) is a system of stimuli and responses in bacterial cells governed by their population density, through which they regulate genes that control virulence factors and biofilm formation. Despite considerable research on QS and the discovery of new antibiotics, QS-controlled biofilm formation by microorganisms in clinical settings has remained a problem because of nascent drug res...
